CagriSema is a novel fixed-ratio co-formulation combining cagrilintide (a long-acting amylin analog) with semaglutide (a GLP-1 receptor agonist) in a single weekly injection. This combination targets two distinct appetite-regulating pathways — amylin and GLP-1 — providing additive weight loss beyond what either agent achieves alone. Phase 3 trials have shown approximately 22-25% body weight reduction, making it one of the most effective anti-obesity medications in development. Cagrilintide enhances satiety through amylin pathways in the brainstem, while semaglutide reduces appetite via GLP-1 receptors, creating a complementary dual mechanism.
